Biological Background: FA2H Function and Localization

  • FA2H, also known as fatty acid alpha-hydroxylase or FAAH, is encoded by the FA2H gene in humans.
  • Catalyzes the C-2 hydroxylation of free fatty acids to produce 2-hydroxy fatty acids, essential precursors for sphingolipids and glycosphingolipids in neural tissue and epidermis. Related references: PMID:15337768, PMID:15863841, PMID:17355976
  • Exhibits stereospecificity for (R)-2-hydroxy fatty acids. Related references: PMID:22517924
  • Plays a critical role in myelin sheath formation by contributing to galactosphingolipid synthesis.
  • Essential for epidermal lamellar body formation and skin permeability barrier function. Related references: PMID:17355976
  • Localizes to the endoplasmic reticulum and microsome membranes.
  • Predominantly expressed in brain and colon, with lower levels in testis, prostate, pancreas, and kidney; also detected in differentiating keratinocytes and epidermis.
  • Contains heme and iron binding sites, belongs to the fatty acid hydroxylase family, and is associated with hereditary spastic paraplegia and leukodystrophy.

Experimental Guidance and Technical Tips

  • The immunogen (aa 95–170) targets a conserved region of FA2H; verify cross-reactivity with mouse and rat orthologs in relevant tissue samples.
  • For Western blot, expect a band near 43 kDa. Use reducing conditions and include positive controls such as brain or colon lysates.
  • In ELISA applications, titrate the antibody against recombinant FA2H or the immunogen peptide to optimize detection sensitivity.
  • As FA2H is a membrane protein, employ lysis buffers containing detergents (e.g., Triton X-100 or SDS) for efficient protein extraction.
