Biological Background: Ezrin Function and Localization

  • Ezrin, also known as Cytovillin, Villin-2, or p81, is encoded by the EZR gene and belongs to the ERM (ezrin-radixin-moesin) protein family.
  • It functions as a linker between the plasma membrane and the actin cytoskeleton, essential for maintaining cell shape and structure.
  • In epithelial cells, Ezrin is required for the formation of microvilli and membrane ruffles on the apical surface.
  • Together with PLEKHG6, it is involved in normal macropinocytosis.
  • Ezrin localizes to the apical cell membrane, microvillus membrane, ruffle membrane, cell cortex, and the cytoskeleton.
  • It is expressed in various tissues including the cerebral cortex, hippocampus, and intestinal epithelial cells, with preferential expression in astrocytes.
  • Post-translational modifications include phosphorylation, acetylation, and S-nitrosylation, which may regulate its activity and interactions.

Experimental Guidance and Technical Tips

  • The antibody was raised against a synthetic peptide within amino acids 300–400 of human Ezrin; consider this epitope location when designing experiments or validating across species.
  • In Western blot analysis, the expected band is around 69 kDa, but variants or modifications may cause slight shifts; validate using appropriate controls.
  • For immunohistochemistry-paraffin (IHC-P), antigen retrieval and permeabilization steps may be necessary since Ezrin is membrane/cytoskeleton-associated; optimize conditions for your sample type.
  • ELISA applications may require testing different coating and detection conditions; this antibody can serve as a detection reagent when paired with a suitable capture antibody.
