Biological Background: EZH2/KMT6 Function and Localization
- Catalyzes mono-, di-, and trimethylation of histone H3 at Lys-27 (H3K27me1/2/3) and methylates Lys-9, with a preference for unmethylated substrates, leading to transcriptional repression. Related references: PMID:14532106, PMID:15225548, PMID:15385962, PMID:15231737, PMID:17210787, PMID:18285464, PMID:22323599, PMID:30923826
- Plays a critical role in embryonic stem cell identity by generating H3K27me3, with higher abundance than EZH1-containing complexes in ESCs. Related references: PMID:19026781
- Serves as a recruitment platform for DNA methyltransferases, linking H3K27 methylation and DNA methylation for coordinated gene silencing. Related references: PMID:16357870, PMID:17200670
- Represses genes including HOXC8, HOXA9, MYT1, CDKN2A, retinoic acid target genes, and circadian clock genes; essential for CRY1/2-mediated repression of PER1/2 transcription (By similarity). Related references: PMID:16179254, PMID:18086877, PMID:20935635, PMID:16717091
- Methylates non-histone substrates such as GATA4 and RORA, expanding its regulatory functions beyond histones. Related references: PMID:23063525
- Primary localization in the nucleus.
- Expressed in many tissues, with specific expression in ovarian follicles; frequently overexpressed in cancers including breast, colon, and lymphoma. Related references: PMID:14532106, PMID:31451685
- Subject to alternative splicing, phosphorylation, glycosylation, and ubiquitin-like conjugation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-250) of human EZH2, which may cross-react with splice variants sharing this region.
- As a polyclonal antibody, expect high sensitivity but validate consistency across production batches.
- For western blot (WB), the predicted molecular weight is ~85 kDa; alternative splicing may yield additional bands.
- Confirm nuclear localization in immunofluorescence/immunocytochemistry (IF/ICC) experiments.
- Given its cross-reactivity with mouse, this antibody is suitable for studies in mouse model systems.
- In ELISA applications, optimize antibody concentration for capture or detection roles based on assay design.
- EZH2 overexpression in many cancers makes it a valuable marker; consider testing in relevant cancer cell lines.
