Biological Background: EXT2 Function and Localization
- Encodes exostosin-2, a type II transmembrane glycosyltransferase that serves as a critical subunit of the heparan sulfate (HS) co-polymerase; alternative names include Glucuronosyl-N-acetylglucosaminyl-proteoglycan 4-alpha-N-acetylglucosaminyltransferase and Multiple exostoses protein 2.
- Forms a heterodimeric complex with EXT1 to constitute the active HS polymerase; together they catalyze the processive elongation of the HS backbone by alternating addition of D-glucuronic acid (GlcA) and N-acetyl-D-glucosamine (GlcNAc) residues from their respective UDP-sugar donors. Related references: PMID:22660413, PMID:36402845, PMID:36593275
- The EXT1 subunit provides the glucuronosyltransferase activity, while EXT2 is responsible for the N-acetylglucosaminyltransferase activity; both are required for full polymerase function and HS chain synthesis. Related references: PMID:36402845, PMID:36593275
- The resulting heparan sulfate proteoglycans are ubiquitous components of the extracellular matrix and cell surfaces, playing essential roles in tissue homeostasis, growth factor signaling, and cell adhesion. Related references: PMID:19344451, PMID:22660413
- Primarily localizes to the Golgi apparatus membrane, with presence in the cis-Golgi network and endoplasmic reticulum membrane; a secreted form has also been reported.
- Widely expressed across tissues.
- Mutations in EXT2 are associated with hereditary multiple exostoses, an autosomal dominant disorder characterized by cartilage-capped bony outgrowths; the protein is also classified as a tumor suppressor.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 80–340) encompasses the catalytic domain of EXT2; the resulting polyclonal antibody is expected to recognize multiple epitopes within this region, providing robust detection for denaturing Western blotting and native applications like IF/ICC.
- For Western blotting, consider a primary antibody dilution range of 1:500–1:2000 and validate against the predicted molecular weight of ~82 kDa; inclusion of appropriate controls (e.g., EXT2-overexpressing lysates or siRNA-treated samples) is recommended.
- In immunofluorescence/ICC, EXT2 typically displays a perinuclear Golgi-like staining pattern; membrane permeabilization with 0.1% Triton X-100 can improve accessibility while preserving organelle morphology.
- ELISA performance is supported by the recombinant protein immunogen; consider using the antibody as a capture or detection reagent in sandwich ELISA setups for HS-related biomarker research.
- Cross-reactivity with mouse and rat EXT2 has been demonstrated, enabling use in rodent models; however, it is advisable to verify specificity in the relevant species and tissue before large-scale studies.
