Biological Background: Exportin-5 Function and Localization
- Exportin-5 (XPO5), also known as Ran-binding protein 21, is a member of the importin-beta family that functions as a nuclear export receptor for double-stranded RNA (dsRNA) and proteins containing dsRNA binding domains (dsRBDs).
- In a RanGTP-dependent manner, XPO5 mediates the nuclear export of micro-RNA precursors (pre-miRNAs) that form short hairpins, a critical step in miRNA biogenesis. Related references: PMID:14631048, PMID:14681208, PMID:15613540
- XPO5 also exports synthetic short hairpin RNAs (shRNAs) used for RNA interference, and can mediate nuclear export of deacylated and aminoacylated tRNAs in some circumstances.
- The protein specifically recognizes dsRNAs lacking a 5'-overhang, with a short 3'-overhang and a double-stranded length of at least 15 base-pairs, in a sequence-independent manner. Related references: PMID:19965479
- During infection, XPO5 mediates the nuclear export of adenovirus VA1 dsRNA, highlighting its role in host-virus interactions.
- XPO5 shuttles between the nucleus and cytoplasm; it loads cargo in the nucleus, docks at the nuclear pore complex via nucleoporins, and releases cargo in the cytoplasm upon RanGTP hydrolysis.
- The protein is widely expressed, with documented presence in heart, brain, placenta, lung, skeletal muscle, kidney, and pancreas.
- Post-translational modifications include acetylation and phosphorylation, and the protein is proteomically identified as a reference proteome component.
Experimental Guidance and Technical Tips
- The immunogen used to generate this antibody corresponds to amino acids 1-200 of human Exportin 5 (XPO5), a region that may be involved in cargo recognition; consider this when assessing binding specificity.
- This antibody is recommended for Western blot (WB), immunohistochemistry on paraffin-embedded sections (IHC-P), and ELISA applications. It is advisable to validate the antibody in the relevant sample system and optimize dilutions empirically.
- Given the predicted molecular weight of 136 kDa, ensure appropriate gel electrophoresis and transfer conditions for high-molecular-weight proteins during WB.
