Biological Background: Exportin 5 Function and Localization
- Exportin-5 (XPO5), also known as Ran-binding protein 21, is a ~136 kDa nuclear transport receptor member of the importin-β superfamily.
- It specifically mediates the nuclear export of double-stranded RNA (dsRNA) and micro-RNA precursors (pre-miRNAs) in a RanGTP-dependent manner (PubMed:14631048, PubMed:14681208, PubMed:15613540). Related references: PMID:14631048 PMID:14681208 PMID:15613540
- XPO5 recognizes dsRNAs with a minimum length of 15 base pairs, a short 3′-overhang, and no 5′-overhang, a structural basis for selective cargo export (PubMed:19965479). Related references: PMID:19965479
- In addition to pre-miRNAs, XPO5 exports viral dsRNAs such as adenovirus VA1, linking it to host-virus interaction and innate immunity.
- The export cycle involves cooperative binding of XPO5 to Ran-GTP and cargo in the nucleus, docking to nucleoporins, and cytoplasmic release upon GTP hydrolysis.
- XPO5 is predominantly localized in the nucleus but shuttles to the cytoplasm, with expression detected in heart, brain, placenta, lung, skeletal muscle, kidney, and pancreas.
- Post-translational modifications include acetylation and phosphorylation, which may regulate its activity or protein interactions.
Experimental Guidance and Technical Tips
- The immunogen spans the C-terminal region (aa 925-1204) of human Exportin 5, which is highly conserved across human, mouse, and rat, supporting cross-reactivity.
- For Western blot, the predicted molecular weight is 136 kDa; use appropriate gel percentage and transfer conditions for high-molecular-weight proteins.
- In immunofluorescence/ICC, expect predominantly nuclear staining with some cytoplasmic localization.
- For IP, use standard protocols with careful optimization of antibody concentration.
- This polyclonal antibody may recognize multiple isoforms or degradation products; validate in the relevant sample system.
