Biological Background: Exportin-2 Function and Localization
- Exportin-2, also known as CAS, CSE1, XPO2, or importin-alpha re-exporter, is encoded by the CSE1L gene.
- Functions as the export receptor for importin-alpha, mediating its re-export from the nucleus to the cytoplasm after cargo release.
- In the nucleus, forms a cooperative trimeric complex with importin-alpha and GTP-bound Ran, docking to nucleoporins for transport through the nuclear pore complex.
- Upon cytoplasmic disassembly, Ran-GTP hydrolysis stimulated by RANBP1 and RANGAP1 releases importin-alpha, allowing Exportin-2 to recycle back to the nucleus.
- The asymmetric distribution of Ran-GTP and Ran-GDP between nucleus and cytoplasm confers directionality to nuclear export.
- Detected in brain, placenta, ovary, testis, and trachea, with high expression in testis and proliferating cells.
