Biological Background: EXOSC7 Function and Localization

  • EXOSC7 (Exosome component 7, also known as RRP42, p8, EAP1) is a non-catalytic subunit of the RNA exosome complex, which possesses 3'->5' exoribonuclease activity.
  • It participates in diverse RNA processing and degradation events in both the nucleus and cytoplasm. In the nucleus, it contributes to the maturation of stable RNAs (rRNA, snRNA, snoRNA) and the elimination of processing by-products and pervasive transcripts.
  • In the cytoplasm, the exosome complex facilitates general mRNA turnover, degrades AU-rich element (ARE)-containing unstable mRNAs, and participates in RNA surveillance to prevent translation of aberrant mRNAs.
  • EXOSC7 is localized to the nucleolus and nucleoplasm within the nucleus, as well as the cytoplasm, enabling its dual role in nuclear and cytoplasmic RNA metabolism.
  • Post-translational modifications include acetylation and phosphorylation, which may regulate its function or interactions within the exosome complex.
  • The RNA exosome, including EXOSC7, is implicated in immunoglobulin class switch recombination and somatic hypermutation by targeting AICDA deamination activity.

  • The immunogen corresponds to the N-terminal region (amino acids 1-100) of human EXOSC7. For Western blot, the predicted molecular weight is approximately 32 kDa. Validate band specificity using appropriate positive and negative controls (e.g., EXOSC7-overexpressing lysates or siRNA knockdown samples).
  • For ELISA, consider using recombinant EXOSC7 protein or cell lysates from human/mouse origin. Titrate the antibody to optimize signal-to-noise ratio.
  • Although validated for WB and ELISA, performance in other applications (e.g., IHC, IF) should be tested experimentally under relevant conditions.
