Biological Background: EXOSC10 Function and Localization
- Catalytic component of the RNA exosome complex, providing 3'->5' exoribonuclease activity for RNA processing and degradation, including rRNA, snRNA, snoRNA maturation and elimination of pervasive transcripts.
- Involved in the SSU processome for pre-rRNA processing and targeted degradation during ribosome biogenesis. Related references: PMID:34516797
- Required for DNA double-strand break repair by homologous recombination: recruits RPA and RAD51, and regulates damage-induced RNAs to limit end resection. Related references: PMID:25632158 PMID:31086179
- Essential for global protein translation and cell proliferation. Related references: PMID:26857222 PMID:36912080
- Regulates metabolism of C9orf72-derived repeat RNA, preventing translation into toxic dipeptide repeat proteins. Related references: PMID:32830871
- Localizes to the cytoplasm and nucleus (nucleolus and nucleoplasm). Undergoes alternative splicing, phosphorylation, and Ubl conjugation.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 586–885 of human EXOSC10, targeting the C-terminal region. Consider designing experiments with this region in mind for epitope masking or competition assays.
- For Western blot, the predicted molecular weight is approximately 101 kDa. Verify band size in your sample system; post-translational modifications may affect migration.
- The antibody is applicable to human, mouse, and rat samples by WB and ELISA. Validate in your specific model, as expression levels may vary.
- As a polyclonal antibody, lot‑to‑lot variability may occur. It is recommended to test each new lot in your assay system.
