Biological Background: ETV4 Function and Localization
- ETV4 (ETS translocation variant 4), also designated PEA3 and E1A-F, is a member of the ETS family of transcription factors, and alternative splice isoforms may be produced.
- It functions as a transcriptional activator, binding to DNA and regulating gene expression in processes such as motor neuron development and keratinocyte differentiation. Related references: PMID:19307308, PMID:31552090
- The protein localizes predominantly to the nucleus, consistent with its role in transcriptional regulation.
- ETV4 is subject to post-translational modifications including phosphorylation and ubiquitination (isopeptide bond), which may modulate its stability and activity.
- During adenovirus infection, ETV4 binds the viral E1A enhancer and acts as a transcriptional activator with a core binding sequence of 5'-[AC]GGA[AT]GT-3'.
- It is required for the precise positioning of motor neurons within the spinal cord and for controlling terminal arborization of motor neuron axons, partly through regulating cadherin and semaphorin expression.
- Expression of ETV4 has been observed in keratinocytes, where it may contribute to epidermal differentiation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 79-178 of human ETV4, an internal region of the protein, which may allow recognition of multiple isoforms if sequence conservation is present.
- For Western blotting, the predicted molecular weight is approximately 54 kDa; consider optimizing blocking and primary antibody dilution for the target species.
- ELISA performance may depend on coating antigen and detection format; titrate the antibody in the relevant sample matrix.
- Cross-reactivity with mouse and rat has been noted, but individual sample validation is recommended when working with these species.
