Biological Background: ETV1 Function and Localization
- ETV1 (ETS translocation variant 1), also known as ER81, is a member of the ETS family of transcription factors that regulate gene expression by binding to specific DNA sequences.
- It functions as a transcriptional activator, recognizing the consensus pentanucleotide 5'-CGGA[AT]-3' (PubMed:7651741). Related references: PMID:7651741
- Plays a critical role in olfactory dopaminergic neuron differentiation, potentially by directly activating the expression of tyrosine hydroxylase (TH).
- Localizes predominantly to the nucleus, consistent with its role as a transcription factor.
- Exhibits tissue-specific expression: very highly expressed in brain; highly in testis, lung, and heart; moderately in spleen, small intestine, pancreas, and colon; weakly in liver, prostate, and thymus; very weakly in skeletal muscle, kidney, and ovary; and not detected in placenta and peripheral blood leukocytes.
- Classified as a proto-oncogene and is associated with chromosomal rearrangements in certain cancers. It undergoes alternative splicing and post-translational modifications including phosphorylation and ubiquitination.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1–200) of human ETV1. For Western blot, consider using nuclear extract preparations from human, mouse, or rat cells/tissues to enrich for the target protein.
- Due to its nuclear localization, sonication or high-salt extraction may improve protein solubilization for WB. Validate antibody specificity in your relevant sample system using appropriate controls.
- ETV1 is highly expressed in brain, testis, lung, and heart; these tissues may serve as positive controls for expression studies.
- For ELISA, ensure that the recombinant immunogen or target protein is properly coated and that antibody detection conditions are optimized.
