Biological Background: Extended Synaptotagmin-1 (ESYT1) Function and Localization
- ESYT1 (Extended synaptotagmin-1), also known as Membrane-bound C2 domain-containing protein, is a calcium-binding protein belonging to the extended synaptotagmin family and contains multiple C2 domains and a transmembrane helix.
- It binds calcium via its C2 domains and translocates to endoplasmic reticulum–plasma membrane contact sites in response to elevated cytosolic calcium, where it helps tether the two organelles and promotes membrane appositions. Related references: PMID:23791178 PMID:24183667
- In the absence of cytosolic calcium, ESYT1 acts as an inhibitor of the ADGRD1 G protein-coupled receptor. Related references: PMID:38758649
- It binds glycerophospholipids via a barrel-like domain and may play a role in cellular lipid transport.
- ESYT1 localizes to both the endoplasmic reticulum membrane and the cell membrane.
