Biological Background: Estrogen Receptor beta Function and Localization
- Serves as a nuclear hormone receptor encoded by the ESR2 gene (also referred to as ER-beta), belonging to the nuclear receptor subfamily 3 group A.
- Binds estrogens with an affinity similar to that of ESR1/ER-alpha and activates expression of estrogen response element (ERE)-containing reporter genes in an estrogen-dependent manner. Related references: PMID:20074560
- Alternative splicing generates multiple isoforms, some of which lack ligand binding ability and exhibit dominant-negative or weakened transcriptional activation.
- Localizes predominantly to the nucleus where it functions as a ligand-activated transcription factor.
- Exhibits tissue-specific expression: detected in testis, ovary, and at lower levels in heart, brain, placenta, liver, skeletal muscle, spleen, thymus, prostate, colon, bone marrow, mammary gland, and uterus; also found in certain tumor cell lines.
- Contains conserved DNA-binding zinc-finger domains and a ligand-binding domain, and is regulated by phosphorylation.
- Plays a role in reproductive development, hormone signaling, and has been implicated in cancer progression.
Experimental Guidance and Technical Tips
- For Western blot (WB): Anticipate multiple bands due to the presence of several isoforms (theoretical sizes include 59, 54, 35, 48, and 42 kDa). Choose the appropriate band based on the tissue and isoform of interest.
- For immunocytochemistry/immunofluorescence (ICC/IF): The antibody detects nuclear-localized ER-beta. Confirm nuclear staining by co-staining with a DNA marker such as DAPI.
- For ELISA: The antibody may be used to detect recombinant or endogenous ER-beta; include appropriate positive and negative controls to validate assay specificity.
- Cross-reactivity: The antibody is reported to recognize ER-beta from human, mouse, and rat. When using other species, perform an initial validation.
- Sample preparation: Use standard protocols for nuclear receptor extraction; consider phosphatase inhibitors as ER-beta is regulated by phosphorylation.
