Biological Background: ESRRB Function and Localization

  • ESRRB encodes the steroid hormone receptor ERR2, also known as ERRβ, estrogen-related receptor beta. It is a member of the nuclear receptor subfamily 3 group B.
  • Functions as a transcription factor by binding to the canonical ESRRB recognition (ERRE) sequence 5'-TCAAGGTCA-3' on target gene promoters and enhancers, regulating their expression. Related references: PMID:17920186, PMID:19755138
  • Plays a critical role in maintaining self-renewal and pluripotency of embryonic and trophoblast stem cells, acting through FGF and Wnt signaling pathways. It is also involved in morula development, particularly at the 8-cell stage.
  • Activates key pluripotency genes such as GATA6, NR0B1, POU5F1, and PERM1. Related references: PMID:23836911
  • Represses transcriptional activities of NFE2L2 and ESR1, providing regulatory crosstalk. Related references: PMID:17920186, PMID:19755138

  • The immunogen is a synthetic peptide spanning amino acids 300–400 of human ESRRB. For Western blot, a band around 48 kDa is expected, although post-translational modifications or isoforms may cause shifts.
  • This antibody is suitable for WB and ELISA. For WB, test a range of dilutions (e.g., 1:500–1:2000) and optimize using positive controls such as embryonic stem cell lysates or tissues known to express ESRRB.
  • Since cross-reactivity with mouse and rat is indicated, this antibody can be used for studies in these species. Validate specificity in the relevant experimental system, preferably with knockdown or knockout controls.
  • For ELISA, consider pairing with a matched detection antibody if used for sandwich assays; the peptide immunogen ensures reactivity with the target protein.
