Biological Background: ESRα (Estrogen Receptor Alpha) Function and Localization
- ESRα, also known as ER-alpha, is a nuclear hormone receptor and a member of the nuclear receptor subfamily 3 group A (NR3A1). It functions as a ligand-activated transcription factor that regulates gene expression in response to estrogen binding.
- Upon estrogen binding, ESRα undergoes conformational changes that promote homodimerization and binding to estrogen response elements (EREs) in target gene promoters, or interaction with other transcription factors such as AP-1, Sp1, and NF-κB, leading to transcriptional activation or repression.
- The receptor plays a critical role in diverse biological processes including cell proliferation, differentiation, and survival in hormone-responsive tissues, and it also mediates rapid membrane-initiated estrogen signaling via kinase cascades.
- Isoforms of ESRα, resulting from alternative promoter usage and splicing, can modulate the transcriptional activity of the full-length receptor through competitive DNA binding or heterodimerization (PubMed:10970861). Related references: PMID:10970861

- For Western blot, endogenous ESRα runs at approximately 66 kDa; consider using appropriate positive and negative cell/tissue controls (e.g., estrogen receptor-positive cell lines) to verify band specificity.
- For ELISA, the polyclonal antibody can be used as a capture or detection reagent; however, optimal dilutions and conditions should be empirically determined.
- Due to the existence of multiple isoforms, confirm the detected band corresponds to the full-length protein or intended isoform in your experimental system.
