Biological Background: ESPN Function and Localization
- ESPN encodes espin, a multifunctional actin-bundling protein also known as autosomal recessive deafness type 36 protein and ectoplasmic specialization protein.
- Plays a major role in regulating the organization, dimension, dynamics, and signaling capacities of actin filament-rich microvilli in mechanosensory and chemosensory cells (PubMed:29572253). Related references: PMID:29572253
- Required for the assembly and stabilization of stereociliary parallel actin bundles and crucial for the formation and maintenance of inner ear hair cell stereocilia.
- Involved in the elongation of actin in stereocilia and in targeting MYO3B to stereocilia tips in extrastriolar hair cells, regulating stereocilia diameter and staircase formation.
- Localizes to the cytoplasm, cytoskeleton, and cell projections including stereocilia and microvilli.
- Contains ANK repeats and a coiled-coil domain; undergoes alternative splicing and phosphorylation.
- Associated with non-syndromic deafness DFNB36, Usher syndrome type I (USH1M), and retinitis pigmentosa.

- When performing IHC on paraffin-embedded sections (IHC-P), optimize antigen retrieval conditions and antibody dilution for your sample type.
