Biological Background: ESE1/ELF3 Function and Localization
- ESE1/ELF3 (ETS-related transcription factor Elf-3), also known as ERT, ESX, EPR-1, ESE-1, or E74-like factor 3, is a member of the ETS transcription factor family. It is encoded by the ELF3 gene and generates multiple isoforms through alternative splicing.
- It functions as a transcriptional activator that binds to ETS sequences containing the consensus core GGA[AT] and transactivates promoters such as collagenase, CCL20, CLND7, FLG, KRT8, NOS2, PTGS2, SPRR2B, TGFBR2, and TGM3.
- ELF3 acts synergistically with POU2F3 to transactivate the SPRR2A promoter and with RUNX1 to transactivate the ANGPT1 promoter, while repressing the KRT4 promoter, highlighting dual regulatory roles.
- The protein is involved in mediating vascular inflammation and serves as a critical downstream effector of the ERBB2 signaling pathway, thereby playing important roles in epithelial cell differentiation and tumorigenesis.
