Biological Background: ERp57 Function and Localization
- ERp57 (also designated PDIA3, GRP58, ERp60, or ERp61) is a member of the protein disulfide isomerase family and functions as an oxidoreductase and molecular chaperone in the endoplasmic reticulum.
- It catalyzes the formation, isomerization, and reduction or oxidation of disulfide bonds in substrate proteins, facilitating correct protein folding. Related references: PMID:11825568, PMID:16193070, PMID:27897272
- ERp57 is a core component of the major histocompatibility complex class I (MHC I) peptide loading complex, where it interacts with TAPBP to assist the assembly of stable MHC I–peptide complexes, playing a critical role in adaptive immunity. Related references: PMID:35948544, PMID:36104323
- Subcellularly, the protein resides primarily in the endoplasmic reticulum lumen and has also been identified in melanosomes.
- At the tissue level, ERp57 is expressed in liver, stomach, and colon, and is detected in gastric parietal cells, chief cells, and the flagellum and head region of spermatozoa. Related references: PMID:20400973, PMID:24188822
- Post‑translational modifications include acetylation, methylation, and phosphorylation; the protein also contains a redox‑active center and disulfide bonds.
Experimental Guidance and Technical Tips
- The immunogen corresponds to residues 206–505 of human ERp57, covering the C‑terminal region that includes the two catalytic thioredoxin‑like domains; this design may allow recognition of both oxidized and reduced enzyme forms.
- For western blotting, a band migrating around 54 kDa is expected under reducing conditions; a reducing sample buffer and 4–20% gradient gel can be used for improved resolution.
- In immunohistochemical (IHC‑P) and immunofluorescence (IF/ICC) applications, optimise antigen retrieval and permeabilization conditions as ERp57 is an ER‑lumenal protein; consider validation in the relevant sample system.
- For ELISA, the antibody can be employed for detection of ERp57 in cell lysates or recombinant protein preparations; perform cross‑validation with other methods to confirm specificity.
