Biological Background: ERLIN1 Function and Localization
- ERLIN1 is a component of the ERLIN1/ERLIN2 complex that mediates endoplasmic reticulum-associated degradation (ERAD) of inositol 1,4,5-trisphosphate receptors (IP3Rs).
- It regulates cellular cholesterol homeostasis by modulating the SREBP signaling pathway and binds cholesterol to promote ER retention of the SCAP-SREBF complex. Related references: PMID:37683630 PMID:24217618
- During hepatitis C virus (HCV) infection, ERLIN1 is required early to initiate RNA replication and later to support infectious virus production.
- It is localized to the endoplasmic reticulum membrane.
- ERLIN1 is expressed in heart, placenta, liver, kidney, pancreas, prostate, testis, ovary, and small intestine.
- Mutations in ERLIN1 are associated with hereditary spastic paraplegia type 62 (SPG62).
- Keywords annotating ERLIN1 include acetylation, cholesterol metabolism, glycoprotein, lipid-binding and neurodegeneration.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a C-terminal region (aa 259-348) of human ERLIN1, which may contribute to specific recognition in WB and IP applications.
- In WB, consider a predicted molecular weight of ~39 kDa; however, endogenous ERLIN1 may appear at a slightly higher molecular weight due to post-translational modifications. Validate in the relevant sample system.
- For IHC-P, expect a membranous staining pattern consistent with ER localization.
- This polyclonal antibody may be suitable for IP; optimize conditions for your experimental setup.
