Biological Background: ERK5 Function and Localization

  • ERK5 (also known as Big MAP kinase 1 (BMK-1) or MAPK7) is a serine/threonine-protein kinase belonging to the MAP kinase family that plays a critical role in cellular proliferation, differentiation, and survival.
  • It is specifically activated by the upstream MAP2K5 (MEK5) kinase, triggering its translocation to the nucleus where it phosphorylates transcription factors such as MEF2C.
  • EGF stimulation activates ERK5 through a Ras-independent, MAP2K5-dependent pathway, distinct from the classic ERK1/2 signaling cascade.
  • In cardiomyocytes, ERK5 acts as a negative regulator of apoptosis by interacting with STUB1/CHIP to promote ubiquitination and degradation of pro-apoptotic ICER isoforms of CREM.
  • ERK5 contributes to endothelial function and blood vessel integrity, and may be involved in muscle cell differentiation.
  • It phosphorylates SGK1 at Ser-78, a modification required for growth factor-induced cell cycle progression.
  • Subcellularly, ERK5 is found in the cytoplasm and nucleus, and concentrates in PML nuclear bodies upon activation.
  • It is expressed in many adult tissues, with highest abundance in heart, placenta, lung, kidney, and skeletal muscle, but is undetectable in liver.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (aa 1-310) of human ERK5, which may detect both full-length and any N-terminally truncated isoforms. Validation in the relevant sample system is advised.
  • For Western blotting, a band near the predicted molecular weight of 88 kDa should be expected; consider using appropriate positive control lysates (e.g., HeLa, HEK293, or tissue samples with high ERK5 expression such as heart or skeletal muscle).
  • In immunohistochemistry (IHC-P) and immunocytochemistry (IF/ICC), optimization of antigen retrieval and permeabilization steps may be required to access nuclear ERK5 pools. Use positive and negative control tissues/cells to confirm staining specificity.
  • For ELISA, whether as capture or detection antibody, cross-reactivity with other MAP kinases should be assessed; testing against recombinant ERK1, ERK2, or JNK is recommended to confirm specificity.
  • Given reported cross-reactivity with mouse and rat, this antibody is suitable for translational studies across these species, but sequence alignment of the immunogen region is recommended to confirm epitope conservation.
