Biological Background: ERK5 Function and Localization
- ERK5, also known as MAPK7, BMK-1, or ERK-5, is a serine/threonine-protein kinase belonging to the MAP kinase family. It plays a central role in the MAPK/ERK signaling cascade, transmitting extracellular signals to regulate diverse cellular processes including proliferation, differentiation, and survival.
- Upon activation by its upstream kinase MAP2K5, ERK5 translocates to the nucleus and phosphorylates downstream targets such as MEF2C. EGF activates ERK5 through a Ras-independent, MAP2K5-dependent pathway, distinguishing it from the classical ERK1/2 modules.
- In cardiomyocytes, ERK5 acts as a negative regulator of apoptosis by interacting with STUB1/CHIP, promoting STUB1-mediated ubiquitination and degradation of ICER-type isoforms of CREM, thereby contributing to cardioprotective signaling.
- ERK5 may be involved in muscle cell differentiation and is important for endothelial function and the maintenance of blood vessel integrity, highlighting its roles in cardiovascular and muscle biology.
- ERK5 phosphorylates SGK1 at Ser-78, a modification required for growth factor-induced cell cycle progression. Additionally, it participates in the regulation of p53/TP53 by disrupting the PML-MDM2 interaction.
- The protein localizes to the cytoplasm, nucleus, and PML nuclear bodies, consistent with its nucleocytoplasmic shuttling upon activation and its involvement in transcriptional regulation.
- Post-translational modifications include acetylation and phosphorylation, which are critical for its kinase activity and signaling functions. ERK5 is expressed in many adult tissues, with high abundance in heart, placenta, lung, kidney, and skeletal muscle, but is not detectable in liver.
