Biological Background: ERK2 Function and Localization
- ERK2 (MAPK1) is a serine/threonine kinase and an essential component of the MAPK/ERK signaling cascade, acting alongside ERK1 (MAPK3) to regulate diverse cellular processes including growth, adhesion, survival, and differentiation through phosphorylation of over 160 substrates. Related references: PMID:35831023
- The kinase phosphorylates a wide array of cytoplasmic, nuclear, and organellar targets—such as transcription factors (ATF2, ELK1, FOS), cytoskeletal proteins (MAP2, CTTN), apoptosis regulators (BAD, MCL1), and translational controllers (EIF4EBP1)—thereby orchestrating gene expression, cytoskeletal dynamics, and apoptosis.
- Under hypoxic conditions, ERK2 phosphorylates PGK1 at the mitochondria to suppress acetyl-coenzyme A formation from pyruvate (PubMed:26942675). Related references: PMID:26942675
- ERK2 also functions as a transcriptional repressor, binding to [GC]AAA[GC] consensus sequences to downregulate interferon gamma-induced genes such as CCL5, IRF7, and OAS1 independently of its kinase activity.
- Subcellular localization is context-dependent: ERK2 is found in the cytoplasm, associated with the cytoskeleton and spindle, in the nucleus, at centrosomes, membrane caveolae, and focal adhesions, reflecting its diverse signaling roles.
- Key post-translational modifications include phosphorylation (activation loop TEY motif), acetylation, and ubiquitination, with implications for signal duration and cellular location. It is classified under keywords: Kinase, Serine/threonine-protein kinase, Phosphoprotein, Acetylation, Ubiquitin conjugation, and others.
Experimental Guidance and Technical Tips
- For Western blotting, the predicted molecular weight is 41 kDa; consider using whole-cell lysates from HeLa, HEK293, or Jurkat cells as positive controls, as ERK2 is ubiquitously expressed.
- In immunohistochemistry (IHC-P), test a range of antigen retrieval methods and antibody dilutions, as ERK2 localization is both nuclear and cytoplasmic; suitable normal and tumor tissue sections should be validated.
- For immunofluorescence (IF/ICC), optimal fixation and permeabilization conditions will preserve both nuclear and cytoskeletal ERK2 signals; consider co-staining with compartment-specific markers.
- The ELISA application may be adapted for quantitative detection; pair with an ERK2 capture antibody and validate in recombinant protein and native sample matrices.
- The immunogen lies within residues 200–300, a region distinct from the regulatory TEY motif; this antibody may recognize both phosphorylated and unphosphorylated ERK2—validate phosphorylation-dependent behavior if needed.
- Since this is a polyclonal antibody, lot-to-lot consistency should be verified in each application; the KO validation (when available) supports target specificity.
