Biological Background: ERK1/2 Function and Localization

  • ERK1 (MAPK3, P27361) and ERK2 (MAPK1, P28482) are the two main mitogen-activated protein kinases, central to the MAPK/ERK signaling cascade that regulates cell proliferation, differentiation, survival, and apoptosis.
  • As serine/threonine kinases, they phosphorylate a vast array of substrates (>160 identified) upon activation, including transcription factors (e.g., ATF2, ELK1, FOS), cytoskeletal proteins, apoptosis regulators, translation regulators, and other kinases and phosphatases, thereby propagating and amplifying the signal.
