Biological Background: eRF3A/GSPT1 Function and Localization
- eRF3A/GSPT1, also known as GST1, ETF3A, eRF3a, or 551G9.2, is a eukaryotic peptide chain release factor GTP-binding subunit (ERF3A) encoded by the GSPT1 gene in humans.
- Functions as a GTPase component of the eRF1-eRF3-GTP ternary complex mediating translation termination at stop codons UAA, UAG, and UGA. Related references: PMID:15987998, PMID:19417105, PMID:2511002
- Mediates delivery of ETF1/ERF1 to stop codons; GTP hydrolysis induces conformational change leading to dissociation of GSPT1/ERF3A, allowing ETF1 to accommodate fully in the ribosomal A-site. Related references: PMID:27863242, PMID:16777602
- Acts as a component of the transient SURF complex that recruits UPF1 to stalled ribosomes, facilitating nonsense-mediated mRNA decay (NMD) of transcripts containing premature stop codons. Related references: PMID:24486019
- Required for SHFL-mediated translation termination that inhibits programmed ribosomal frameshifting (-1PRF) in both viral and cellular genes. Related references: PMID:30682371
- Keywords associated with GSPT1 include GTP-binding, hydrolase activity, nucleotide-binding, protein biosynthesis, alternative splicing, and nonsense-mediated mRNA decay.
- Predicted molecular weight is approximately 56 kDa (calculated 55.7 kDa), consistent with the UniProt annotation.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to the N-terminal region (amino acids 1–100) of human eRF3A/GSPT1 (NP_001123479.1), which may confer specificity to the N-terminus of the protein.
- Given the broad reactivity (Human, Rat), validate the antibody in relevant sample systems using appropriate controls, especially if using for species not explicitly listed.
- Expected band size in Western blot is ~56 kDa; minor bands of different sizes may correspond to isoforms generated by alternative splicing.
- For immunofluorescence or immunohistochemistry, consider optimizing fixation and permeabilization conditions, as the target is predominantly cytoplasmic.
- This product is validated for WB, IF/ICC, IF-P, IHC-P, and ELISA; ensure individual protocol optimization for each application.
