- DNA excision repair protein ERCC-1 (ERCC1), also known as UV20, COFS4, and RAD10, is a non-catalytic component of a structure-specific DNA repair endonuclease.
- It is responsible for the 5'-incision during DNA repair and plays a critical role in the repair of interstrand cross-links (ICL) in conjunction with SLX4.
- ERCC1 participates in processing anaphase bridge-generating DNA structures, which arise from incompletely processed DNA lesions during S or G2 phase and can lead to cytokinesis failure if unresolved.
- The protein is also required for homology-directed repair (HDR) of DNA double-strand breaks, working with SLX4.
- Notably, ERCC1 is not functional in the nucleotide excision repair (NER) pathway, indicating a specialized role in other repair mechanisms.
- It localizes primarily to the nucleus but can also be found in the cytoplasm.
- Post-translational modifications include acetylation and ubiquitination (isopeptide bond formation), and the protein undergoes alternative splicing.
