Biological Background: Erbin Function and Localization
- Erbin (also known as Densin-180-like protein, Erbb2-interacting protein, Protein LAP2) is encoded by the ERBIN (ERBB2IP) gene and exists in alternatively spliced isoforms.
- Erbin functions as an adaptor protein for the receptor tyrosine kinase ERBB2 in epithelial cells, binding to the unphosphorylated Tyr-1248 residue and potentially stabilizing the receptor's inactive state. Related references: PMID:16203728
- Erbin inhibits NOD2-dependent NF-κB signaling and suppresses pro-inflammatory cytokine secretion. Related references: PMID:16203728
- In keratinocytes, Erbin regulates differentiation by binding and sequestering SHOC2, preventing its participation in RAS-RAF-ERK signaling complexes. Related references: PMID:23524970
- Localizes to multiple cellular compartments: cell junctions (hemidesmosomes), basolateral cell membrane, cytoplasm, and nucleus membrane.
- Highly expressed in brain, heart, kidney, muscle, and stomach; lower levels in liver, spleen, and intestine.
- Contains leucine-rich repeats; is a phosphoprotein with multiple modification sites.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1152–1371 within the C-terminal region of human Erbin, which may be involved in protein–protein interactions; consider this when interpreting detection patterns.
- For Western blotting, the predicted molecular weight is ~158 kDa; optimize gel percentage (e.g., 6–8% SDS-PAGE) and transfer conditions accordingly.
- Validate antibody specificity using positive (e.g., human epithelial cell lysates) and negative controls; inclusion of a blocking peptide or recombinant protein can confirm target-specific bands.
- In ELISA, the recombinant immunogen can serve as a positive control to establish assay sensitivity and linearity.
- The antibody is polyclonal and may recognize multiple isoforms or degradation fragments; confirm expected band patterns in your experimental system.
- Although validated for WB and ELISA, cross-reactivity with other family members or off-target proteins should be assessed if applied to other techniques.
