Biological Background: EPS8 Function and Localization
- EPS8 (Epidermal growth factor receptor kinase substrate 8) is a signaling adapter protein that regulates actin cytoskeleton dynamics, controlling processes like cell migration and dendritic cell function. It was initially identified as a substrate for the EGF receptor kinase.
- The protein functions as part of a trimeric complex with SOS1 and ABI1 to transduce signals from Ras to Rac by activating Rac-specific GEF activity, linking growth factor signaling to actin remodeling.
- EPS8 exhibits dual actin-modulating activities: barbed-end capping when bound to ABI1 (inhibiting motility) and actin bundling when associated with BAIAP2 to promote filopodial extensions and membrane protrusions.
- In neurons, EPS8 negatively regulates axonal filopodia formation through actin-capping; however, BDNF-induced phosphorylation inhibits this capping activity, enabling filopodial growth.
- At stereocilia tips in hair cells, EPS8 forms a complex with WHRN and MYO15A that is essential for elongation of the actin core, and mutations in EPS8 are associated with non-syndromic deafness (DFNB102).
- Subcellularly, localizes to the cell cortex, ruffles, growth cones, stereocilia, and synaptosomes, consistent with its roles in dynamic actin-driven membrane protrusions.
- The protein contains an SH3 domain and undergoes phosphorylation and ubiquitination; ubiquitin-mediated degradation is required during G2 phase for cell shape changes associated with cell cycle progression.
- Widely expressed across tissues including brain, heart, lung, liver, kidney, and pancreas, as well as in many epithelial and fibroblastic cell lines.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-200) of human EPS8, which includes part of the SH3 domain and N-terminal region. This region is relatively conserved across human, mouse, and rat, explaining the cross-reactivity. Consider sequence alignment if working with other species.
- For Western blot, the predicted molecular weight is ~92 kDa (based on UniProt: 91.9 kDa). Bands may be observed near this size; however, phosphorylation or other modifications could cause slight shifts. Validate with appropriate positive and negative controls.
- In ELISA applications, employ well-characterized recombinant EPS8 protein or cell lysates from lines with known expression. Since EPS8 is widely expressed, choose a system with moderate to high expression levels.
- As a polyclonal antibody, lot-to-lot consistency may vary; consider performing pilot experiments when switching lots, particularly for quantitative assays.
