  • It functions as a serine protease inhibitor critical for male fertility, modulating the hydrolysis of semenogelin-1 (SEMG1) by prostate-specific antigen (PSA/KLK3). EPPIN binds SEMG1, inhibiting sperm motility and providing antimicrobial protection in the ejaculate coagulum.
  • EPPIN is localized on the cell surface and secreted, covering the surface of testicular and ejaculated spermatozoa. It is expressed by Sertoli cells in the testis and epithelial cells in the epididymis.
  • The protein exhibits alternative splicing, producing isoform 2 which is expressed specifically in the epididymis.
