- EPPIN (Epididymal protease inhibitor), also known as cancer/testis antigen 71 (CT71), is a member of the WAP four-disulfide core domain family (WFDC7) and contains both Kunitz and WAP domains.
- It functions as a serine protease inhibitor critical for male fertility, modulating the hydrolysis of semenogelin-1 (SEMG1) by prostate-specific antigen (PSA/KLK3). EPPIN binds SEMG1, inhibiting sperm motility and providing antimicrobial protection in the ejaculate coagulum.
- EPPIN is localized on the cell surface and secreted, covering the surface of testicular and ejaculated spermatozoa. It is expressed by Sertoli cells in the testis and epithelial cells in the epididymis.
- The protein exhibits alternative splicing, producing isoform 2 which is expressed specifically in the epididymis.
- Key structural features include multiple disulfide bonds, characteristic of the WAP domain, contributing to its stability and function.
- EPPIN possesses antimicrobial activity, protecting spermatozoa within the ejaculate coagulum.

- For Western blotting, endogenous EPPIN is detected at approximately 15 kDa; consider optimizing sample preparation from testis or epididymis tissue lysates.
- For ELISA applications, validate the antibody in direct or sandwich formats using recombinant EPPIN or biological fluids such as seminal plasma.
- Due to the secreted nature of EPPIN, cell culture supernatants from Sertoli or epididymal cell lines may be suitable sources for detection.
