Biological Background: Ephrin A1 Function and Localization

  • Ephrin-A1 is a GPI-anchored cell surface ligand that binds to Eph receptor tyrosine kinases, mediating contact-dependent bidirectional signaling crucial for cell migration, repulsion, and adhesion during neuronal, vascular, and epithelial development.   • It plays a key role in angiogenesis by regulating vascular endothelial cell differentiation and migration through activation of EPHA2.
  • Acts as a ligand for EPHA3, inhibiting epithelial-to-mesenchymal transition of cardiac cells and contributing to heart development. Related references: PMID:12794130
  • May also contribute to dendritic spine morphogenesis (by similarity).
  • Subcellular localization: anchored to the cell membrane via a GPI moiety; also found as a secreted form.

  • The immunization peptide (amino acids 1–100) targets the N-terminal extracellular region, which may improve recognition of both cell surface and secreted ephrin-A1 forms. Consider this when designing capture or detection assays.
  • In Western blot (WB), the predicted protein weight is ~24 kDa. Due to glycosylation and GPI-anchor processing, the actual band may appear at a higher molecular weight. Validate in relevant human or mouse tissue/cell lysates.
  • For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), the membrane and secreted nature of ephrin-A1 may require optimized fixation and permeabilization conditions to preserve antigenicity. Test with known positive controls (e.g., brain tissue, endothelial cells).
  • In ELISA, this monoclonal antibody can serve as a capture or detection reagent. Pair with another clone or a polyclonal antibody targeting a distinct epitope for sandwich ELISA development.
