Biological Background: EPHB4 Function and Localization

  • Encodes a receptor tyrosine kinase that binds transmembrane ephrin-B ligands, mediating contact-dependent bidirectional signaling (forward and reverse signaling) between adjacent cells.
  • Together with its cognate ligand EFNB2, regulates cell adhesion, migration, and repulsion, playing essential roles in heart morphogenesis, angiogenesis, and blood vessel remodeling.
  • Phosphorylates inactive tyrosine-protein kinase EPHB6, promoting its interaction with SH2 domain-containing signaling adapters. Related references: PMID:34431498
  • Localized to the cell membrane, consistent with its function as a transmembrane receptor.
  • Abundantly expressed in placenta, with detectable levels in kidney, liver, lung, pancreas, skeletal muscle, heart, and certain hematopoietic cells.
  • Associated with angiogenesis, developmental processes, and disease variants (as indicated by keywords such as 'Disease variant' and 'Developmental protein').

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 300–540, a region encompassing part of the extracellular domain, which may contribute to specificity for native EPHB4. When using this antibody, consider validating recognition of the target in the relevant sample system.
  • For WB applications, prepare lysates from cell lines or tissues known to express EPHB4 (e.g., placenta, kidney, or liver) and expect a band near 108 kDa. Include appropriate controls to confirm specificity.
  • For ELISA, the antibody can be explored as a capture or detection reagent; pairing with an antibody targeting a non-overlapping epitope may improve sandwich assay performance.
  • Given cross-reactivity with mouse, validate the antibody in mouse-derived samples if intended for such models; note that the immunogen design is based on human sequence.
