Biological Background: EPHB2 Function and Localization
- The EPHB2 gene encodes Ephrin type-B receptor 2, a receptor tyrosine kinase also known as DRT, EK5, ERK, HEK5, Tyro5, and EPHT3. It belongs to the ephrin receptor subfamily of the protein-tyrosine kinase family.
- EPHB2 binds to transmembrane ephrin-B ligands on adjacent cells, triggering bidirectional signaling that regulates axon guidance during development. It is critical for commissural axon pathfinding, contralateral inner ear efferent targeting, and retinal ganglion cell axon navigation to the optic disk.
- In addition to axon guidance, EPHB2 controls dendritic spine development and maturation, and promotes excitatory synapse formation. Activation by EFNB1 reverses ARHGEF15-mediated suppression of synaptogenesis.
- EPHB2 participates in angiogenesis, palate development, and inner ear development by regulating endolymph production. Forward and reverse signaling through the EFNB2/EPHB2 complex also coordinate cell movements during urethral tubularization and cloacal septation.

- Subcellularly, EPHB2 localizes to the cell membrane, axon projections, dendritic processes, and can be secreted. It is widely expressed, with high levels in fetal brain and adult brain, heart, lung, kidney, placenta, pancreas, liver, and skeletal muscle.
- Key post-translational modifications include phosphorylation, glycosylation, disulfide bonds, and isopeptide bond formation. The protein undergoes alternative splicing and ubiquitination.
Experimental Guidance and Technical Tips
- The immunogen comprises amino acids 250–540, which spans part of the extracellular domain and the transmembrane region. This design may allow recognition of both full-length membrane-bound and secreted forms of EPHB2.
- For Western blot (WB), consider using whole-cell lysates from human or mouse brain tissue, given the high expression of EPHB2 in the nervous system. Validate detection under reducing and non-reducing conditions, as disulfide bonds are present.
- For ELISA, the polyclonal nature may provide strong signal but consider potential cross-reactivity with related ephrin receptor family members. Include appropriate controls to confirm specificity.
