Biological Background: EPHA4 Function and Localization

  • Ephrin type-A receptor 4 (EPHA4, also known as EK8, SEK, HEK8, TYRO1) is a member of the Eph receptor family of receptor tyrosine kinases, encoded by the EPHA4 gene.
  • EPHA4 binds membrane-anchored ephrin ligands on adjacent cells, initiating bidirectional signaling (forward through the receptor and reverse through the ligand). It is uniquely promiscuous, activated by both GPI-linked ephrin-A ligands (e.g., EFNA1) and transmembrane ephrin-B ligands (e.g., EFNB3).
  • In the developing nervous system, EPHA4 plays a critical role in axonal guidance, including the establishment of corticospinal projections, and segregates motor and sensory axons during neuromuscular circuit formation.
  • Beyond development, EPHA4 regulates synaptic plasticity in the adult brain and inhibits axonal regeneration after injury. It also contributes to central nervous system vascular formation.
  • Subcellularly, EPHA4 localizes to the cell membrane, axonal and dendritic projections, postsynaptic density membranes, early endosomes, and adherens junctions.
  • EPHA4 is ubiquitously expressed across tissues and participates in diverse cell–cell signaling events, including thymic epithelium development and cochlear pillar cell separation. It can phosphorylate CAPRIN1 to promote formation of membraneless compartments (By similarity).

Experimental Guidance and Technical Tips

  • The immunogen spans the N‑terminal extracellular domain (aa 20–100), thus the antibody is expected to recognize the full‑length receptor and may detect soluble fragments or cleaved ectodomain.
  • In WB using reducing conditions, anticipate a band migrating at ~110 kDa in lysates from human, mouse, or rat tissues/cell lines. Run appropriate positive and negative controls for specificity validation.
  • For IF/ICC, EPHA4 typically exhibits plasma membrane and neuronal projection staining. Use optimized fixation (e.g., 4% PFA) and permeabilization to preserve epitope accessibility and morphology.
  • In ELISA applications, the extracellular epitope recognition can be exploited for sandwich assays or ligand‑binding competition studies. Validate the assay with recombinant EPHA4 protein if available.
  • Because EPHA4 is highly conserved, cross‑reactivity with other Eph receptors cannot be excluded; identity of detected bands should be confirmed by siRNA knockdown or KO models when possible.
