Biological Background: EpCAM Function and Localization

  • EpCAM (Epithelial cell adhesion molecule), also known as TROP1, EGP40, and CD326, is a transmembrane glycoprotein encoded by the EPCAM gene.
  • Functions as a homophilic cell adhesion molecule, mediating interactions between intestinal epithelial cells and intraepithelial lymphocytes, contributing to the mucosal immunological barrier.
  • Plays a role in embryonic stem cell proliferation and differentiation; up-regulates the expression of FABP5, MYC, and cyclins A and E.
  • Localizes to the lateral cell membrane and tight junctions, contributing to cell-cell adhesion and junctional integrity.
  • Highly and selectively expressed on undifferentiated embryonic stem cells, with levels rapidly diminishing upon differentiation.
  • Expressed in almost all epithelial cell membranes, but absent from mesodermal and neural cell membranes; prominently found on the surface of adenocarcinomas.
  • Implicated in hereditary nonpolyposis colorectal cancer (Lynch syndrome) and recognized as a tumor antigen; contains a signal peptide and disulfide bonds.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal extracellular domain (amino acids 1–100) of human EpCAM, encompassing a region critical for homophilic interactions.
  • In Western blotting (WB), a band of approximately 35 kDa is expected; however, native glycosylated EpCAM may migrate at a higher molecular weight. Validate detection in epithelial cell lines (e.g., MCF-7, HepG2) which express EpCAM endogenously.
  • For immunohistochemistry-paraffin (IHC-P), EpCAM is strongly expressed on the membrane of epithelial cells. Consider appropriate antigen retrieval methods and include positive (e.g., colon carcinoma) and negative (e.g., mesenchymal tissue) controls.
  • In ELISA applications, this monoclonal antibody can be tested as a capture or detection reagent for quantifying soluble or cell-surface EpCAM. Cross-reactivity with mouse and rat EpCAM may facilitate analysis across species; however, epitope conservation should be verified.
