- Epcam (Epithelial cell adhesion molecule), also known as EGP314, CD326, TROP1, and Tacsd1, is a type I transmembrane glycoprotein involved in cell-cell adhesion.
- It contains an N-terminal signal peptide, an extracellular domain with thyroglobulin type-1-like repeats, a single transmembrane helix, and a short cytoplasmic tail.
- Epcam localizes to the lateral cell membrane and tight junctions in epithelial cells, where it mediates homophilic interactions between intestinal epithelial cells and intraepithelial lymphocytes, contributing to the mucosal immunological barrier.
- The protein plays a role in embryonic stem cell proliferation and differentiation, and it up-regulates the expression of FABP5, MYC, and cyclins A and E.
- It is a disulfide-linked glycoprotein, and its expression is associated with various cancers, often referred to as Tumor-associated calcium signal transducer 1.

- For Western blot (WB), consider using whole-cell lysates from epithelial tissues or cell lines (e.g., mouse colon or human Caco-2 cells), and validate the band at the expected molecular weight of ~35 kDa.
- In ELISA applications, the antibody may be used to capture or detect Epcam in solution; however, optimal coating concentrations and blocking conditions should be empirically determined.
