Biological Background: ENTPD5 Function and Localization
- ENTPD5, also known as CD39L4, NTPDase-5, or PCPH, is an endoplasmic reticulum-resident nucleoside diphosphatase belonging to the ectonucleoside triphosphate diphosphohydrolase family.
- The enzyme preferentially hydrolyzes nucleoside diphosphates such as GDP, IDP, and UDP, with lower activity toward ADP and CDP. Related references: PMID:10400613 PMID:15698960
- In the ER lumen, ENTPD5 clears UDP, a feedback inhibitor of UDP-Glc:glycoprotein glucosyltransferases, thus positively regulating protein reglucosylation, a process critical for glycoprotein folding and quality control.
- Subcellular localization is predominantly the endoplasmic reticulum; however, secretion has also been reported.
- Tissue expression is highest in adult liver, kidney, prostate, testis, and colon, with much weaker expression in other tissues.
- Key features include calcium/magnesium-dependent hydrolase activity, proto-oncogene classification (PCPH), disulfide bond formation, and glycosylation, highlighting roles in cellular signaling and cancer biology.
