Biological Background: ENTPD1/CD39 Function and Localization
- ENTPD1, also known as CD39, is an ectonucleoside triphosphate diphosphohydrolase 1 (NTPDase1) belonging to the ENTPD family.
- It catalyzes the hydrolysis of nucleoside triphosphates (NTPs) and diphosphates (NDPs) in a sequential two-step reaction, converting NTPs to nucleoside monophosphates (NMPs) via NDP intermediates. Related references: PMID:8529670, PMID:8626624, PMID:8955160
- This activity regulates extracellular nucleotide levels; by hydrolyzing proinflammatory ATP and platelet-activating ADP, it blocks platelet aggregation and supports blood flow. Related references: PMID:8955160, PMID:8996251
- The protein localizes to the plasma membrane and caveolae, consistent with its function as an ecto-enzyme.

- When using immunohistochemistry (IHC-P) or immunofluorescence (IF/ICC, IF-P), consider antigen retrieval methods and validate with appropriate controls, given the membrane-bound localization of the target.
