Biological Background: ENSA Function and Localization
- Alpha-endosulfine (ENSA), also known as ARPP-19e, is a protein phosphatase inhibitor that specifically inhibits protein phosphatase 2A (PP2A) during mitosis.
- When phosphorylated at Ser-67 during mitosis, ENSA interacts with the regulatory subunit PPP2R2D (PR55-delta), leading to PP2A inactivation, which is essential to maintain high cyclin-B1-CDK1 activity during M phase (By similarity).
- ENSA also acts as a stimulator of insulin secretion by interacting with the sulfonylurea receptor (ABCC8), thereby preventing sulfonylurea binding and reducing K(ATP) channel currents.
- Localizes to the cytoplasm.
- Widely expressed with high levels in skeletal muscle and brain and lower levels in the pancreas.
- Post-translational modifications include phosphorylation and acetylation; involved in cell cycle, mitosis, and protein phosphatase inhibition.
- Alternative splicing yields multiple isoforms.
