Biological Background: ENPP3/CD203c Function and Localization
- ENPP3, also known as CD203c, B10, NPP3, PDNP3, and PD-Ibeta, is a member of the ectonucleotide pyrophosphatase/phosphodiesterase family. It functions as a hydrolase that metabolizes extracellular nucleotides, including ATP, GTP, UTP, and CTP. Related references: PMID:29717535 PMID:9344668
- It plays a critical role in innate immune regulation by hydrolyzing 2',3'-cGAMP, a STING agonist, thereby modulating type-I interferon production. Related references: PMID:38749434
- ENPP3 limits mast cell and basophil responses during allergic inflammation by eliminating extracellular ATP, a signaling molecule that activates these cells in an autocrine manner.
- In the small intestine, it metabolizes extracellular ATP to prevent ATP-induced apoptosis of intestinal plasmacytoid dendritic cells.
- The enzyme also hydrolyzes dinucleoside polyphosphates, including vasoactive adenosine polyphosphates. Related references: PMID:12846830
- Subcellularly, ENPP3 is located at the cell membrane, with enrichment at the apical cell membrane; a secreted form is also detected.
- Tissue specificity: detected on bile ducts in liver, in blood serum, prostate, uterus, and on basophils. Related references: PMID:15072822 PMID:9344668 PMID:11342463
- It is a calcium- and zinc-binding metalloenzyme with multiple disulfide bonds, a transmembrane helix, and a signal-anchor sequence.
Experimental Guidance and Technical Tips
- For Western blotting, consider that the predicted molecular weight of ENPP3 is approximately 100 kDa; validate with appropriate gel percentage and transfer conditions.
- In immunofluorescence or immunocytochemistry, expect membranous and possibly apical staining patterns; confirm localization in relevant human cell lines or tissues.
- The antibody is suitable for flow cytometry on live cells due to the epitope region (amino acids 551–875) being extracellular; consider gating strategies for basophil or mast cell populations.
- For ELISA, the antibody can be used to detect recombinant or native ENPP3; include appropriate positive and negative controls.
