Biological Background: ENPP3 Function and Localization
- ENPP3 (Ectonucleotide pyrophosphatase/phosphodiesterase family member 3), also known as CD203c, NPP3, PDNP3, and PD-IBETA, is a type II transmembrane glycoprotein and member of the ENPP family of extracellular nucleotide hydrolases.
- As a nucleotide pyrophosphatase/phosphodiesterase, ENPP3 hydrolyzes extracellular nucleotides, including ATP, GTP, UTP, and CTP, thereby regulating purinergic signaling and extracellular nucleotide pools. Related references: PMID:29717535, PMID:9344668
- ENPP3 can also degrade 2',3'-cGAMP, a second messenger that activates the STING pathway, thus acting as a negative regulator of type I interferon production and innate immune responses. Related references: PMID:38749434
- The enzyme limits mast cell and basophil activation during inflammation and chronic allergic responses by eliminating extracellular ATP, a key autocrine signaling molecule for these cells.
- Additionally, ENPP3 hydrolyzes dinucleoside polyphosphates, including vasoactive adenosine polyphosphates, suggesting a role in vascular tone regulation. Related references: PMID:12846830

Experimental Guidance and Technical Tips
- The immunogen, a recombinant fusion protein spanning amino acids 646–875 of human ENPP3, corresponds to the C-terminal extracellular domain. This design may enable detection of both full-length transmembrane and shed soluble forms of ENPP3.
- For Western blot (WB), a band around 100 kDa is expected; consider validating the antibody in relevant human, mouse, or rat tissue lysates or cell lines known to express ENPP3 (e.g., basophils, prostate, liver).
- In ELISA applications, the antibody can be used either as a detection or capture reagent. Optimization of coating concentrations and detection conditions is recommended for best sensitivity.
- Cross-reactivity with mouse and rat ENPP3 is reported, but sequence homology in the immunogen region should be verified for these species; testing in the target species’ samples is advised.
