Biological Background: ENO3 Function and Localization
- Beta-enolase (ENO3) is a muscle-specific enolase that catalyzes the conversion of 2-phosphoglycerate to phosphoenolpyruvate in glycolysis and the reverse reaction in gluconeogenesis.
- It is alternatively named 2-phospho-D-glycerate hydro-lyase, muscle-specific enolase (MSE), and skeletal muscle enolase, and is encoded by the ENO3 gene.
- ENO3 plays a critical role in striated muscle development and regeneration, with expression primarily in skeletal and cardiac muscle.
- In striated muscle, ENO3 forms functional homodimers (β/β) or heterodimers with alpha-enolase (α/β), while in most other tissues alpha/alpha homodimers predominate.
- The protein localizes to the cytoplasm and is subject to post-translational modifications including acetylation and phosphorylation.
- Alternative splicing produces multiple isoforms, and mutations in ENO3 are associated with glycogen storage disease type 13 (GSD13).
- ENO3 requires magnesium as a cofactor and is classified as a lyase involved in metal-ion binding and glycolytic processes.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1-190 of human ENO3 (NP_001967.3), covering the N-terminal region. Consider that this region may exhibit conservation across isoforms and species when interpreting cross-reactivity data.
- For Western blot (WB), a predominant band at approximately 47 kDa is expected. Validation of signal specificity with appropriate controls (e.g., knockdown or tissue-specific expression) is recommended.
- This polyclonal antibody is reported for use in WB, IF/ICC, and ELISA. For applications beyond those listed, titration and thorough validation in the relevant sample system are advised.
