Biological Background: Alpha-enolase Function and Localization
- Alpha-enolase catalyzes the conversion of 2-phosphoglycerate to phosphoenolpyruvate in glycolysis and the reverse reaction in gluconeogenesis. Related references: PMID:1369209 PMID:29775581
- Eno1 is involved in various processes such as growth control, hypoxia tolerance and allergic responses. Related references: PMID:10802057 PMID:12666133 PMID:2005901
- It functions as a receptor and activator of plasminogen on the cell surface of several cell-types such as leukocytes and neurons, contributing to the intravascular and pericellular fibrinolytic system. Related references: PMID:12666133
- Stimulates immunoglobulin production. Related references: PMID:1369209
- Acts as a transcriptional repressor by binding to the myc promoter and may function as a tumor suppressor.
- Localizes to the cytoplasm, cell membrane, nucleus, and specifically to the M line of sarcomeres in myofibrils.
- The alpha/alpha homodimer is expressed in embryo and in most adult tissues; the alpha/beta and beta/beta homodimers are found in striated muscle, and the alpha/gamma and gamma/gamma homodimers in neurons.
- Post-translationally modified by acetylation, phosphorylation, hydroxylation, and ubiquitin-like conjugation; requires magnesium as a cofactor.
Experimental Guidance and Technical Tips
- The immunogen covers the full-length human ENO1 sequence (aa 1–434), which may allow recognition of multiple epitopes.
- For Western blotting, a band at approximately 47 kDa is expected; validate with appropriate positive and negative controls.
- In immunofluorescence or immunocytochemistry, anticipate cytoplasmic and membrane staining; nuclear localization may also be observable in certain contexts.
- For immunoprecipitation, antibody concentration and incubation conditions should be optimized for efficient capture.
- ELISA performance can be verified using the recombinant immunogen as a standard.
- Cross-reactivity with mouse and rat ENO1 has been demonstrated; confirm in your specific experimental system.
