Biological Background: EMR2 Function and Localization

  • EMR2, also known as ADGRE2 or CD312, belongs to the adhesion G protein-coupled receptor (aGPCR) family and contains characteristic EGF-like repeats in its extracellular domain.
  • Functions as a cell surface receptor for glycosaminoglycans, specifically binding chondroitin sulfate moieties to mediate cell attachment.
  • Promotes innate immune responses by inducing granulocyte chemotaxis, degranulation, and adhesion.
  • In macrophages, EMR2 activation triggers the release of pro-inflammatory cytokines IL-8 and TNF.
  • Serves as a regulator of mast cell degranulation. Related references: PMID:26841242
  • Localizes to the plasma membrane and is enriched in membrane ruffles, consistent with its role in cell motility and adhesion.
  • Exhibits myeloid-restricted expression, with high levels in peripheral blood leukocytes, mast cells, and myeloid cell lines, and is absent from non-immune tissues.

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 300–500 of human EMR2, covering part of the extracellular EGF-like domain; it may recognize both full-length and autocatalytically cleaved receptor forms.
  • For Western blotting, expect a band around 90 kDa; note that extensive glycosylation can cause the observed molecular weight to be higher.
  • For ELISA, the antibody may be used as capture or detection reagent; titration and optimization with recombinant EMR2 protein are recommended.
  • Reported cross-reactivity with mouse EMR2 supports translational studies in murine models; validate with appropriate tissue or cell line controls.
