Biological Background: EMC4 Function and Localization
- EMC4, also known as PIG17 or TMEM85, is a subunit of the endoplasmic reticulum membrane protein complex (EMC), a multi-protein insertase that facilitates the biogenesis of membrane proteins.
- The EMC mediates energy-independent insertion of newly synthesized membrane proteins into the ER membrane, with a preference for substrates containing weakly hydrophobic or destabilizing transmembrane domains. Related references: PMID:29242231, PMID:29809151, PMID:30415835
- It is essential for the cotranslational insertion of multi-pass membrane proteins, ensuring proper stop-transfer membrane-anchor sequence integration into ER membranes. Related references: PMID:29809151, PMID:30415835
- Additionally, EMC4 is required for the post-translational insertion of tail-anchored (TA) proteins into the ER membrane. Related references: PMID:29242231, PMID:29809151
- By mediating proper cotranslational insertion of N-terminal transmembrane domains in an N-exo topology, the EMC controls the topological orientation of multi-pass membrane proteins, such as G protein-coupled receptors (GPCRs). Related references: PMID:30415835
- EMC4 localizes to the endoplasmic reticulum membrane. Alternative splicing generates isoforms with distinct tissue expression—isoform 1 is expressed in brain and heart, while isoform 2 is expressed in heart.
- The protein is subject to acetylation and phosphorylation, and has been identified in proteomics analyses; it is annotated with keywords including alternative splicing, apoptosis, and transmembrane domains.
Experimental Guidance and Technical Tips
- Immunogen design: The recombinant immunogen covers amino acids 1–100 of human EMC4 (N-terminal region). Consider verifying antibody reactivity against the full-length endogenous protein, as this region may include both structured and disordered elements.
- Western blot: The antibody detects bands around 15 kDa, 16 kDa, and 20 kDa, likely reflecting post-translational modifications or isoform differences. Validate these patterns in your specific sample system.
- IHC-P: Antigen retrieval and detection conditions should be optimized for each tissue type. No single protocol is universally applicable; pilot studies are recommended.
- ELISA: Suitable for standard assay formats; however, coating conditions and detection ranges must be empirically determined.
- Cross-reactivity: The antibody cross-reacts with mouse and rat EMC4, but optimal working dilutions may vary between species. Titration is advised.
- General note: All experiments should include appropriate positive and negative controls to confirm specificity.
