Biological Background: Very Long Chain Fatty Acid Elongase 5 Function and Localization

  • ELOVL5 encodes the very long chain fatty acid elongase 5, a key enzyme in the elongation cycle of long-chain fatty acids, and multiple isoforms are produced through alternative splicing.
  • It catalyzes the first and rate-limiting reaction of the long-chain fatty acids elongation cycle in the endoplasmic reticulum, adding two carbons per cycle to long- and very long-chain fatty acids (VLCFAs).
    Related references: PMID:10970790 PMID:20937905
  • The enzyme acts as a condensing enzyme with high specificity toward polyunsaturated acyl-CoA, particularly C18:3(n-6) acyl-CoA, and is involved in producing monounsaturated and polyunsaturated VLCFAs that serve as precursors of membrane lipids and lipid mediators.
    Related references: PMID:10970790 PMID:20937905
  • ELOVL5 is localized primarily to the endoplasmic reticulum membrane and is also found in cell projections such as dendrites.
  • Ubiquitously expressed, it shows high expression in the adrenal gland and testis, with weaker levels in prostate, lung, and brain, and is detected in the cerebellum.
  • In the absence of essential linoleic and alpha-linolenic acids, ELOVL5 participates in the synthesis of Mead acid from oleic acid, an alternative pathway for polyunsaturated fatty acid production.
  • Mutations in ELOVL5 are associated with spinocerebellar ataxia type 38 (SCA38), a neurodegenerative disorder, highlighting its critical role in neural lipid metabolism.
  • Post-translational modifications include acetylation and phosphorylation, which may regulate its activity and function.

Experimental Guidance and Technical Tips

  • For Western blot, consider using whole cell lysates from tissues with high ELOVL5 expression such as adrenal gland or testis. The predicted molecular weight is approximately 35kDa; optimize gel percentage and transfer conditions accordingly.
  • For ELISA, the polyclonal nature may provide broad reactivity; validate the antibody using recombinant ELOVL5 protein or native samples and include appropriate negative controls.
  • The immunogen corresponds to amino acids 247–326 in the C-terminal region; verify reactivity with specific isoforms if alternative splicing is a concern in your experimental system.
  • As a rabbit polyclonal antibody, confirm consistency across different batches for reproducibility in long-term studies.
