Biological Background: ELMO1 Function and Localization
- ELMO1 (Engulfment and cell motility protein 1), also known as ced-12 homolog, is a key regulator of DOCK family guanine nucleotide exchange factors (GEFs), including DOCK1, DOCK2, and DOCK5. Related references: PMID:11595183, PMID:12134158, PMID:32651375
- In complex with DOCK and CRK, ELMO1 promotes RAC1 activation, a critical step in cytoskeletal rearrangements required for phagocytosis of apoptotic cells and cell motility. Related references: PMID:11595183, PMID:38857861
- ELMO1 is widely expressed across tissues, with elevated expression in the spleen and placenta, consistent with its role in immune cell function.
- The protein localizes to the cytoplasm and cell membrane, where it participates in signal transduction and cytoskeletal reorganization.
- ELMO1 is subject to post-translational modifications including phosphorylation and acetylation, and contains SH3-binding motifs that mediate protein-protein interactions.
- Alternative splicing of ELMO1 generates multiple isoforms with varying tissue distribution and functional properties.
Experimental Guidance and Technical Tips
- For Western blotting, use a starting sample load of 20–40 µg of total protein lysate from human or mouse cells/tissues, as ELMO1 is an 84 kDa protein with variable expression levels.
- Validate the antibody in your specific sample system and application by including appropriate positive and negative controls; consider using siRNA knockdown or knockout cell lines to confirm band specificity.
- For ELISA applications, this polyclonal antibody may serve as a capture or detection reagent; optimize coating and detection concentrations based on assay format.
- The immunogen corresponds to the C-terminal region (aa 527–727), which may recognize multiple ELMO1 isoforms; note that potential cross-reactivity with ELMO family members (ELMO2, ELMO3) should be evaluated if relevant.
