Biological Background: ELL Function and Localization
- ELL (Eleven-nineteen lysine-rich leukemia protein) is an RNA polymerase II elongation factor, also known as MEN, ELL1, and PPP1R68, encoded by the ELL gene at chromosomal band 19q13.2.
- It functions as an elongation factor component of the super elongation complex (SEC) and the little elongation complex (LEC). The SEC increases RNA polymerase II transcription elongation by suppressing transient pausing, whereas LEC regulates small nuclear RNA (snRNA) gene transcription. - Prior to SEC assembly, ELL helps stabilize RNA polymerase II recruitment/initiation and facilitates entry into the pause site.
- Subcellularly, ELL localizes to the nucleus, with enrichment in nuclear speckles and Cajal bodies.
- It is ubiquitously expressed, with highest levels in placenta, skeletal muscle, testis, and peripheral blood leukocytes.
- ELL undergoes post-translational acetylation and phosphorylation, and chromosomal rearrangements involving ELL are associated with leukemia, designating it a proto-oncogene.
Experimental Guidance and Technical Tips
- The immunogen peptide corresponds to amino acids 350–450 of human ELL; this region is conserved, allowing cross-reactivity with mouse ELL.
- For Western blot, a predominant band at approximately 68 kDa is expected in whole-cell or nuclear extracts from human and mouse cells. Validate with loading controls and consider using nuclear fractionation to enrich the signal.
- For ELISA, immobilize the immunogen peptide or recombinant ELL protein as the capture antigen and optimize antibody dilution to achieve linear detection.
- Potential batch-to-batch variation is inherent to polyclonal antibodies; therefore, titrate each new lot for the intended application.
