Biological Background: ELF5 Function and Localization
- ELF5 (E74-like factor 5), also known as ESE-2, is an ETS family transcription factor encoded by the ELF5 gene.
- It acts as a transcriptional activator and may regulate later stages of keratinocyte terminal differentiation.
- Isoform 2 binds to DNA sequences with the core motif GGA[AT], and activates the SPRR2A and PSP gene promoters.
- ELF5 is localized to the nucleus, consistent with its role as a transcription factor.
- Expression is restricted to epithelial-rich tissues, including salivary gland, mammary gland, kidney, and prostate; expression is upregulated during keratinocyte differentiation.
- Multiple isoforms exist due to alternative splicing, with isoform 1 and isoform 2 showing differential tissue expression; in kidney only isoform 1 is expressed, while prostate expresses both.
- Several epithelial carcinoma cell lines show lack of ELF5 expression, suggesting potential tumor-suppressive or regulatory roles.
- Keywords: Activator, Alternative splicing, DNA-binding, Nucleus, Transcription regulation.
Experimental Guidance and Technical Tips
- Given the immunogen spans amino acids 40-150, the polyclonal antibody may recognize multiple ELF5 isoforms if the epitope is conserved. For Western blotting, a band around 31 kDa is expected; validate using human, mouse, or rat epithelial cell lysates.
- For immunohistochemistry (IHC-P), normal salivary gland or mammary gland tissues serve as positive controls; expect nuclear staining. Optimize antigen retrieval and antibody dilution empirically.
- For immunofluorescence (IF/ICC), confirm nuclear localization and consider co-staining with epithelial markers. The antibody’s cross-reactivity with mouse and rat enables use in rodent models, but dilutions must be validated per experimental system.
- In ELISA, use recombinant ELF5 protein or cell lysates as capture antigens; proper positive and negative controls should be included.
