Biological Background: ELF4 Function and Localization
- ELF4 (E74-like factor 4), also referred to as myeloid Elf-1-like factor (MEF), is an ETS-related transcription factor encoded by the ELF4 gene (Gene ID: 2000).
- It functions as a transcriptional activator that binds to DNA sequences containing the consensus 5'-WGGA-3' motif, regulating genes involved in hematopoiesis and immune responses.
- ELF4 transactivates promoters of hematopoietic growth factors, including CSF2 (GM-CSF), IL3, and IL8, and acts synergistically with RUNX1 to activate IL3 expression.
- It plays a critical role in the development and function of natural killer (NK) cells, NK T-cells, and CD8+ T-cells, controlling CD8+ T-cell proliferation and homing via KLF4 and KLF2.
- ELF4 orchestrates inflammatory responses by sustaining the transcription of anti-inflammatory genes (e.g., IL1RN) and negatively regulating pro-inflammatory cytokines such as IL17A, IL1B, IL6, TNFA, and CXCL1. Related references: PMID:34326534 PMID:35266071

- Abundantly expressed in placenta and myeloid leukemia cell lines, with moderate expression in heart, lung, spleen, thymus, peripheral blood lymphocytes, ovary, and colon; lower levels in T-cell lines and absent in brain.
