Biological Background: ELF2 Function and Localization
- ELF2 (E74-like factor 2), also designated EU32 and NERF (New ETS-related factor), is a transcription factor encoded by the ELF2 gene.
- Two main isoforms exist: isoform 1 transcriptionally activates the LYN and BLK promoters, acting synergistically with RUNX1; isoform 2 may function in repression of RUNX1-mediated transactivation.
- As a member of the ETS (E26 transformation-specific) family, ELF2 contains a conserved ETS DNA-binding domain that recognizes purine-rich sequences.
- ELF2 is localized to the nucleus, consistent with its role in transcriptional regulation.
- The protein is subject to multiple post-translational modifications, including phosphorylation, methylation, and ubiquitination (isopeptide bond formation), which may modulate its activity and stability.
- ELF2 exhibits broad tissue expression; in fetal tissues, highest levels in heart, lung, liver, and kidney; in adult tissues, prominent in heart, placenta, lung, skeletal muscle, spleen, thymus, testis, and ovary.

- The immunogen corresponds to amino acids 1–140 of human ELF2 (N-terminal region); thus, the antibody may recognize both major isoforms.
- For Western blot (WB), a band around 64 kDa is expected; consider using nuclear-enriched lysates from human or mouse cells/tissues with known ELF2 expression.
- For ELISA applications, the antibody can be utilized as either capture or detection reagent; preliminary titration and cross-reactivity testing against the target and closely related ETS family members are recommended.
