Biological Background: ELAVL4 Function and Localization
- ELAVL4 (ELAV-like protein 4), also known as HuD or paraneoplastic encephalomyelitis antigen HuD, is encoded by the ELAVL4 gene and belongs to the ELAV/Hu family of RNA-binding proteins.
- It plays a central role in post-transcriptional regulation by binding to AU-rich elements (AREs) in the 3' UTR of target mRNAs, stabilizing them and protecting from decay; targets include GAP43, VEGF, FOS, CDKN1A, and ACHE. Related references: PMID:10710437, PMID:12034726, PMID:12468554
- Additionally, ELAVL4 regulates alternative splicing, as shown for CALCA pre-mRNA, and modulates translation in a poly(A)- and cap-dependent manner. Related references: PMID:17035636, PMID:10710437
- It is essential for neuronal development, promoting differentiation of neural progenitor cells, neurite outgrowth, dendrite maturation, and synapse function, and contributes to learning and memory. Related references: PMID:12034726, PMID:12468554, PMID:17234598
- Subcellularly, ELAVL4 localizes to the cytoplasm, perikaryon, dendrites, axons, and growth cones, aligning with its roles in mRNA transport and local protein synthesis.
- Expression is predominantly detected in the brain and pancreatic beta cells at the protein level.
- Post-translational modifications include methylation and phosphorylation; it is recognized as the HuD autoantigen in paraneoplastic encephalomyelitis.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the N-terminal domain (aa 1–100), so epitopes may be accessible in both native and denaturing conditions; consider antigen retrieval for IHC-P and fixation optimization for IF-P.
- This polyclonal antibody can detect endogenous ELAVL4 in human, mouse, and rat samples; validate specificity with knockout controls or peptide blocking where possible.
- For WB, a band near 42 kDa is expected; multiple isoforms or post-translational modifications may produce additional bands.
- Given its role in RNA biology, RNA-binding assays (e.g., RIP) may benefit from UV crosslinking prior to immunoprecipitation.
